Which security headers matter most?

Strict-Transport-Security (HSTS), Content-Security-Policy, X-Content-Type-Options and a frame-ancestors or X-Frame-Options rule protect against the most common attacks.

Can I check internal or localhost URLs?

No. For safety, only public websites on the standard ports 80 and 443 can be checked.
